10% CO2 incubator

RPMI 1640 + Glutamax
FBS 10%
Penicillin/streptomycin 1%

Mix RPMI + P/S, let ON in the incubator (cap unscrewed), then add the FBS the next day

TPA 200nM (always added fresh on the cells)

Only after thawing the cells:

Add PTU at 200 µM,

After 2-3 days change with medium without PTU, and let the cells grow and become darker for a least a week.

Max passage P20 + P10

Split every 2-3 days, 2 in 5 or 1 in 2
