Biochemical and Infection Characteristics of Escherichia coli O157:H7-specific Bacteriophages and Broad-host-range Bacteriophages for E. coli Serotypes
The objective of the present study was to analyze biochemical and infectious characteristics of 7 E. coli O157:H7 specific bacteriophages (narrow coliphage group, NCG) and 7 broad-host-range bacteriophages for E. coli serotypes (broad coliphage group, BCG) out of 55 coliphages. There were no big differences on burst sizes of 50-176 PFU/infected cell and harborring virulence factors for stx1, stx2, ehxA, and saa. However, the results revealed that NCG might be more stable than BCG under harsh acidic pH and high ethanol. All the NCG infected only to 23 E. coli O157, but BCG showed broad infection spectrum for E. coli O157:H7 and other 61 E. coli. All the NCG belonged to the Siphoviridae of 50 kb genome size and all the BCG were Myoviridae of 130 kb genome size. As the host-phage adsorption characteristics, all the NCG infected E. coli O157:H7, but not E. coli O157:H7△rfaL, deletion mutant of O-antigen ligase gene for lipopolysaccharide. Five coliphages among NCG infected E. coli O103, but not E. coli O103△rfaG, deletion mutant of the glycosyltransferase gene. NCG of Siphoviridae recognized O-antigen of E. coli O157, but BCG of Myoviridae might recognize O-antigen and/or a part of the lipopolysaccharide core as an adsorption site in various E. coli. The receptor of NCG and BCG interacts with some part of lipopolysaccharide, and the tail morphology of the coliphages might be related to their adsorption to and recognition of a different part of lipopolysaccharide. Therefore, E. coli O157:H7 specific bacteriophages showed very different characteristics of high stabilities, morphology of Siphoviridae, and host-adsorption site on the O-antigen lipopolysaccharide of E. coli O157:H7.
